RARβ Expression in Keratinocytes from Potentially Malignant Oral Lesions: The Functional Consequences of Re-Expression by De-Methylating Agents

چکیده
Loss of RARβ2 expression by promoter methylation is an early event in oral carcinogenesis. Understanding the mechanisms and consequences RARβ loss may aid understanding disappointing results retinoid chemoprevention trials. This study aimed to describe effects all-trans retinoic acid (ATRA) de-methylating agent 5-Aza-2′ deoxycytidine (5-AZA-CdR) on a panel immortal potentially malignant lesion (PMOL) cell cultures. was assessed PMOL tissues immunohistochemistry. Cells were treated with ATRA ± 5-AZA-CdR, cycle senescence assessed. In tissues, variable, but lower biopsies which gave rise Treatment iPMOL cells resulted little change expression, addition 5-AZA-CdR significant increases. The variable be related as this has wider cycle. Overall, response treatment dependent several factors, including RARβ-promoter methylation. These findings help explain lack consistent effect retinoids PMOLs seen

epithelial expression of keratinocytes growth factor in oral precancer lesions
background: keratinocyte growth factor (kgf) is a potent epithelial mitogen that acts by binding the kgf receptors (kgfrs) expressed on epithelial cells and regulates proliferation and differentiation. the objective of this study was to investigate the expression of kgf in the epithelium in oral precancer. materials and methods: archival tissues of oral submucous fibrosis (smf) and leukoplakia ...
متن کاملExpression of ABCG2 and Bmi-1 in oral potentially malignant lesions and oral squamous cell carcinoma
Early diagnosis is vital for effective treatment of oral squamous cell carcinoma (OSCC). The optimal time for clinical intervention is prior to malignancy when patients present with oral potentially malignant lesions such as leukoplakia or erythroplakia. Transformation rates for oral dysplasia vary greatly and more rigorous methods are needed to predict the malignant potential of oral lesions. ...
متن کاملIncreased nuclear ?-catenin expression in oral potentially malignant lesions: A marker of epithelial dysplasia
BACKGROUND Deregulation of β-catenin is associated with malignant transformation; however, its relationship with potentially malignant and malignant oral processes is not fully understood. The aim of this study was to determine and compare the nuclear β-catenin expression in oral dysplasia and oral squamous cell carcinoma (OSCC). MATERIAL AND METHODS Cross sectional study. Immunodetection of ...
متن کاملPotentially malignant oral lesions: clinicopathological correlations
OBJECTIVE To determine the incidence of potentially malignant oral lesions, and evaluate and correlate their clinical and pathological aspects. METHODS The sample consisted of cases clinically diagnosed as oral leukoplakia, oral erythroplakia, erythroleukoplakia, actinic cheilitis, and oral lichen planus treated at a diagnostic center, between May 2012 and July 2013. Statistical tests were co...
متن کاملEx vivo confocal imaging with contrast agents for the detection of oral potentially malignant lesions.
OBJECTIVES We investigated the potential use of real-time confocal microscopy in the non-invasive detection of occult oral potentially malignant lesions. Our objectives were to select the best fluorescence contrast agent for cellular morphology enhancement, to build an atlas of confocal microscopic images of normal human oral mucosa, and to determine the accuracy of confocal microscopy to recog...
